The main image showing the diatoms was doing using oblique lighting (Olympus Aplanat Achromat condenser) with 450nm LED light and a monochrome converted Nikon d850 camera with a 2.5x Nikon CF PL photoeyepiece.
The cross polarized images were done using white LED light and a Canon Eos 5DSR camera (auto whitebalance). A linear polarizer was placed on the field lens and another on top of the photoeyepiece. An Olympus Aplanat Achromat condenser provided the lighting.
Strong crystallization of the quinidine is seen across the slide (as often occurs with these types of slides), although different parts of the slide provide different crystal growths. In the diatom image the crystals can be seen as white structures in the background. In the cross polarized image with the two large crystal growths, diatoms can be seen as dark features within the crystalline regions.
