A strew slide which is mainly Amphora ovalis. Prepared by Samuel Henry Meakin. Mounted in Styrax. Olympus BHB microscope using 450nm LED light. 63x Leitz Pl Apo 1.4 objective, oil immersion. Olympus Aplanat Achromat condenser, oil immersion, oblique lighting. 2.5x Nikon CF PL photoeyepiece. Monochrome converted Nikon d850 camera. 32 images stacked in Zerene (Pmax).
There are some other diatoms on the slide. I selected one, a Navicula sp. of some type, and did a single image with the 63x objective and oblique lighting. This one was nice and flat and a lot of the areolae were in focus at the same time. Measurement in ImageJ showed the areolae spacing to be 323nm.
